why does 5-HT₂C antagonism produce selective overeating?  Optogenetic and pharmacologic studies in mice (2023–2024) showed that 5-HT₂C antagonism specifically inhibits POMC neurons in the hypothalamus and activates AgRP/NPY neurons (appetite stimulators).  In Cell Reports (2024), a study was published showing that direct injection of a selective 5-HT₂C antagonist into the nucleus accumbens increases consumption of highly-palatable food without altering homeostatic hunger; i.e., this pathway primarily enhances hedonic (pleasure-seeking) drive for food, not merely energy need.  Another study combining chemogenetic silencing and metabolic radio-telemetry (2024) revealed that part of the weight gain resulting from 5-HT₂C antagonism also originates from a reduction in basal energy expenditure (via inhibition of brown adipose tissue), not only from overeating.
